Abstract

Chronic wounds (principally pressure sores, venous leg ulcers and diabetic foot ulcers) are a drain on global health services and remain a major area of unmet clinical need. Chronic wounds are characterised by a stable and stubborn bacterial biofilm (densely aggregated colonies of bacteria encased by a matrix of extracellular polymeric substances (EPS)) which hinders innate immune response and delays or prevents wound healing. Bioactive glass (BG) fibres doped with antimicrobial metal ions can offer a promising treatment for chronic wound infections.
In this thesis, firstly, an existing partial differential equation (PDE) model of a chronic wound infection is explored and adapted to investigate the combined antibacterial and wound healing effects of metal-doped BG fibres. From solutions, it was observed that the bacterial infection must be eradicated for a chronic wound to heal successfully. Accord- ingly, the development and characterisation of BG fibres doped with the antimicrobial ions, silver and copper, was next considered to investigate their antibacterial/anti-biofilm properties.
Anti-biofilm studies using the bacteria Pseudomonas aeruginosa & Staphylococcus au- reus demonstrated that treatment efficacy depended on the species used. Additionally, treatment was more effective against single-species biofilms compared to dual-species.
